1.A PCR reaction produces an amplicon (DNA strand) of 766bp which contains a single restriction enzyme cut site for EcoRI and for BamHI.You perform a single digestion with each restriction enzyme and a double digestion using both enzymes and separate the products on a 2% agarose gel with a 50 bp DNA sizing ladder.


You know that the EcoRI digestion yields fragments of 425 bp and 341 bp and BamHI yields 700 bp and 66 bp in the 5’ to 3’ direction. Indicate in the gel diagram below where the bands would be and explain your reasoning.The ladder is already inserted for you.

t DNA has a net negative charge therefore, it moves towards the positive terminal of the gel when loaded in the gel for Agarose Gel Electrophoresis (AGE). Upon PCR, multiple copies of this 766 base pair (bp) amplicon will be produced. Uncut fragment of 766 bp will be observed more closely towards the well and in between the mark of 700 and 800 bp of ladder.
